Cell culture Bone marrow–derived macrophages (BMDM) from WT and Apoe mice were prepared as adherent cultures using recombinant mouse M-CSF (30 ng/ml, Peprotech). For ectopic expression of apoE in Apoe BMDMs and Ly-6C monocytes, the cells were cultured in Opti-

Mice All mice were housed and bred in specific pathogen–free conditions in the Animal Research Facility at the San Francisco Veterans Affairs Medical Center. All animal experiments were approved by the Institutional Animal Care and Use Committee at the VA Medical Center. WT C57BL/6, Ldlr (B6.129S7-Ldlr/J) and Apoe (B6.129P2-Apoe/J) mice (3-4 weeks old) were purchased from The Jackson Laboratory and fed with a rodent chow diet containing 4.0% fat (Harlan Teklad, 2916), or high fat diet (21% fat, 1.5% cholesterol, Research Diets, D12079B). ApoeLdlr and ApoeLdlr mice, established as previously described, were fed a rodent chow diet of equal nutritional value containing 4.0% fat (Harlan Teklad, 2916) and 9.0% fat (Harlan Teklad, 2919) respectively.

Onset of apoprotein E secretion during differentiation of mouse bone marrow-derived mononuclear phagocytes
A number of macrophage functions were sequentially expressed when the bone marrow precursors of mononuclear phagocytes differentiated in culture in the presence of a specific growth factor, colony-stimulating factor-1. We have defined the expression of apoprotein E (ApoE), a major secreted protein of resident peritoneal macrophages, during maturation of adherent bone marrow-derived mononuclear ...
متن کاملInducible ApoE gene repair in hypomorphic ApoE mice deficient in the low-density lipoprotein receptor promotes atheroma stabilization with a human-like lipoprotein profile.
OBJECTIVE To study atherosclerosis regression in mice after plasma lipid reduction to moderately elevated apolipoprotein B (apoB)-lipoprotein levels. APPROACH AND RESULTS Chow-fed hypomorphic Apoe mice deficient in low-density lipoprotein receptor expression (Apoe(h/h)Ldlr(-/-)Mx1-cre mice) develop hyperlipidemia and atherosclerosis. These mice were studied before and after inducible cre-medi...
متن کاملMacrophage phospholipid transfer protein deficiency and ApoE secretion: impact on mouse plasma cholesterol levels and atherosclerosis.
OBJECTIVE PLTP and apoE play important roles in lipoprotein metabolism and atherosclerosis. It is known that formation of macrophage-derived foam cells (which highly express PLTP and apoE) is the critical step in the process of atherosclerosis. We investigated the relationship between PLTP and apoE in macrophages and the atherogenic relevance in a mouse model. METHODS AND RESULTS We transplan...
متن کاملLow-Density Lipoprotein Receptor–Related Protein 1 Prevents Early Atherosclerosis by Limiting Lesional Apoptosis and Inflammatory Ly-6C Monocytosis Evidence That the Effects Are Not Apolipoprotein E Dependent
absence of apoE and LRP1 promoted atherogenesis more than did macrophage apoE deletion alone in both apoE-producing LDLR / mice ( 88%) and apoE / mice ( 163%). The lesions of both mouse models with apoE / LRP1 / macrophages had increased macrophage content. In vitro, apoE and LRP1 additively inhibit macrophage apoptosis. Furthermore, there was excessive accumulation of apoptotic cells in lesion...
متن کاملHyperhomocysteinemia promotes inflammatory monocyte generation and accelerates atherosclerosis in transgenic cystathionine beta-synthase-deficient mice.
BACKGROUND Hyperhomocysteinemia (HHcy) is an independent risk factor for cardiovascular disease. Monocytes display inflammatory and resident subsets and commit to specific functions in atherogenesis. In this study, we examined the hypothesis that HHcy modulates monocyte heterogeneity and leads to atherosclerosis. METHODS AND RESULTS We established a novel atherosclerosis-susceptible mouse mod...
